What the clinical trials actually used.

Shafiee et al. (2024) in Nutrition Reviews pooled 8 randomized controlled trials comparing saffron head-to-head with SSRIs. No significant difference in depression or anxiety outcomes, with fewer adverse events in the saffron group. Every trial in the pool used standardized extract at roughly 80-100mg a day over 6-8 weeks.

Earlier meta-analyses by Hausenblas (2013) and Lopresti and Drummond (2014) found the same pattern: saffron beat placebo for mood at a real dose of verified extract. But saffron is one of the most counterfeited spices on earth, so the dose on the label and the testing behind it, not the word "saffron" on the front, are what separate a product that works from colored powder.

Several small randomized trials found standardized saffron extract was associated with improvements in low mood versus placebo at around 80 to 100mg a day. Those trials reached that total with 28 to 30mg capsules taken two or three times daily, which is why many supplements use that per-capsule amount. One capsule alone is roughly a third of the studied dose. These are small trials, and saffron is a supplement, not a prescription.

Saffron is one of the most adulterated spices in the world, so this matters. Look for published third-party testing, crocin and safranal levels on the label, and GMP certification. Products without those can contain safflower, turmeric, or dye. If a saffron supplement is suspiciously cheap, treat that as a warning sign.
It builds slowly, not the same day. The studies started recording a difference around 2 to 4 weeks of daily use, with the fuller effect by 6 to 8 weeks. If you feel nothing after a couple of months of a full, verified dose, it likely isn't the right fit for you.
